Methods of designing or modifying proteinstructure at the protein or genetic level to producespecified amino-termini in vivo or in vitro aredescribed. The methods can be used to alter themetabolic stability and other properties of theprotein or, alternatively, to artificially generateauthentic amino-termini in proteins produced throughartificial means, The methods are based upon theintroduction of the use of artificial ubiquitin-protein fusions, and the discovery that the in vivohalf-life of a protein is a function of the amino-terminal amino acid of the protein.
