(57) [Abstract] The present invention provides a variety of nucleic acid using repair enzymes. The present invention provides a method for detecting point mutations in nucleic acid sequences. The present invention further provides a method of detecting the wild type nucleic acid sequence or nucleic acid sequence having no mutation. The present invention, be joined to nucleic acid repair enzyme to the probe molecule, by and used tail labeling techniques and (oscillation reaction) and (tail labeling technique) oscillation reaction, it is intended to enhance the detection of the target polynucleotide. The present invention provides a helical promoting molecules and (helix destabillization molecule) spiral unregulated molecules to enhance the hybridization of the target polynucleotide of the probe. The present invention also provides use of thermostable nucleic acid repair enzyme to facilitate the reaction at elevated temperatures. The present invention provides a method for measuring the index of repair of an oligonucleotide probe with a mismatched or damaged. Finally, the present invention can be induced effectively in vitro mutagenesis of a target polynucleotide.
