Recovery of Mn peroxidases (MnP) comprises adding an Mn-containing nutrient liquid to an Agaricales white rot fungus in an open vessel where the fungus is fixed to a carrier prior to the nutrient liquid being added and the added liquid is subsequently removed from the vessel after a definite reaction period.TF - TECHNOLOGY FOCUS - BIOLOGY - Preferred Process: The fungus is a pure Agricales culture, e.g. Clitocybula duseni (DSM 11679, 07.08.1997), Nematoloma frowardi (DSM 11680, 07.08.1997) or Stropharia rugosoannulata or their Mn peroxidase-forming variants or mutants. The nutrient liquid is a Mn-containing succinate-/phosphate-buffered liquid or Mn chloride and has a Mn concentration of above 100 mum. It is removed with the carried fungus from the vessel after less than 2 weeks (especially after 2-6 days) and is subjected to ultrafiltration. For further Mn-P recovery in a semi-continuous operation the removed nutrient liquid can be replaced and to interrupt the MnP recovery in batch operation the fixed fungus is stored between operations at below 10degreesC without the added liquid nutrient. In order to increase the enzyme yield the nutrient liquid is stirred in contact with the carried fungus.
