Microbial production of amino acids (A) of the asparate and/or glutamate families comprises increasing pyruvate carboxylase (PC) activity by genetic alteration of the enzyme and/or increasing expression of the PC gene in appropriate microorganisms. Independent claims are also included for the following: (1) a PC gene (I) encoding a 1104 amino acid enzyme (II) and/or its allelic variants; (2) gene construct containing (I); (3) vector containing (I) or the construct; and (4) transformed cells containing, in a replicable form, (I) or the construct.
