PROBLEM TO BE SOLVED: To provide the subject novel gene that comprises the flagellin gene of Clostridium chauvoei having a specific base sequence and is useful for vaccine of for blackleg being a communicable acute fever disease infecting domestic animals mainly ruminants and for standard and inspection of diagnosis or diagnostic solution. ;SOLUTION: This is a novel flagellin gene of Clostridium chauvoei having the base sequence described in the formula and is useful for vaccine, diagnosis and the standard and inspection of the diagnostic solution of blackleg, a communicable acute fever disease infecting domestic animals, particularly ruminants. The flagellin gene is obtained by culturing Clostridium chauvoei ATCC 10092, recovering the cell bodies, extracting the chromosomal DNA by breaking cell walls of the cell bodies, partially cleaving the DNA with restriction enzymes into fragments, packaging these fragments into λ-phage, allowing the λ-phage to infect to Escherichia coil, screening the formed plaque with a polyclonal antibody specific to the Clostridium chauvoei flagellums and recovering DNA from the positive clones.;COPYRIGHT: (C)1999,JPO
