PROBLEM TO BE SOLVED: To obtain a new DNA fragment which contains toluene monooxygenase gene having a specific restriction enzyme map, and can be used for producing an enzyme useful for the remediation of polluted environments by degrading an aromatic compound, a halogenated aliphatic hydrocarbon, and so on. SOLUTION: This is a new DNA fragment, having a size of about 5.3 kb, which contains toluene monooxygenase gene having numbers of cleavable sites by restriction enzymes of 3 (BamHI), 1 (clal), 1 (EcoRI), 3 (KpnI), 2 (NcoI), 2 (NspV), 2 (ScaI), 2 (SmaI), 2 (SphI), 1 (StuI), 0 (DraI), 0 (EcoRV), 0 (HindIII), 0 (HpaI), 0 (NdeI), 0 (PvuII), 0 (SacI), 0 (Sse8387I), 0 (XbaI), and 0 (XhoI), and has a restriction enzyme map shown by the formula. A transformant containing it is useful for producing an enzyme useful for the remediation of polluted environments by degrading an aromatic compound, a halogenated aliphatic hydrocarbon, and so on. This fragment is obtained from Ralstonia eutropha strain TB64.
