摘要

The present invention relates to genetic engineering.More specifically, a method is presented for reducing mispriming duringDNA synthesis. In particular, the present invention relates to universalprimers which reduce mispriming during cDNA library construction,thereby increasing the proportion of cDNA clones having been primedfrom the bona fide 3' poly A tail. The present invention further relates tothe use of the discriminating oligonucleotides of the present invention inother methods such as mRNA purification, PCR-based detection methodsand sequencing.
