SUBSTANCE: invention relates to determined mutant modified thermolysine-like neutral metalloprotease with amino acid sequence of "wild" type where leucine-144 is substituted for serine, or aspartic acid-150 is substituted for asparagine, histidine or lysine, or glutamic acid-187 is substituted for glutamine, or asparagine-227 is substituted for histidine, lysine, arginine, or leucine-144 is substituted for serine, or aspartic acid-150 is substituted for histidine and asparagine-227 is substituted for histidine, or leucine-144 is substituted for serine, or aspartic acid-150 is substituted for asparagine and asparagine- -227 is substituted for histidine. Obtained enzymes are used in methods of synthesis and hydrolysis of benzylhydroxycarbonyl- --L-aspartyl-L-phenylalanine methyl ester that involves interaction of corresponding substrate solutions with modified thermolysine-like neutral proteases.
