Particularly a 30 thy A strain of Vibrio Cholerae that lacks thy A functionality is disclosed. This strain can comprise one or more episomic elements that autonomously replicate DNA, such as plasmids that have a functional gene in thy A, optionally foreign, for example, E. coli that allows the strain to develop in the absence of thymine in the development medium and which optionally has a structural gene encoding a homologous or heterologous protein. In addition, proteins encoded by a structural thy A gene and the 5 'flanking region are described as SEQ ID NO: 4 in Figure 4 and SEQ ID NO: 5 in Figure 5, respectively. Additionally a vaccine comprising a 30 thy A strain of Vibrio cholerae of the invention or a thy A strain of Vibrio cholerae produced by the process of the invention is disclosed.
