PURPOSE: A catabolite repression resistant mutant strain is provided from thermophilic Bacillus thermoglucosidasius by distorting a repression mechanism of enzyme production and main glucose absorption mechanism of bacteria artificially. CONSTITUTION: A catabolite repression resistant mutant is prepared by treating thermophilic Bacillus thermoglucosidasius with mutagens such as NTG(N-methyl-N'-nitro-N-nitroso guanidine) and UV; screening a mutant strain in the present of 2-deoxyglucose; culturing the mutant strain in a nutrient rich agar medium, which contains 0.28-0.32% of glucose, 0.9-1.0% of starch or cellulose, at 52-58C for 22-26 hours; dyeing plate medium with 0.1N of iodine or Congo-Red reagent then isolating the catabolite repression resistance mutant strain by collecting the population forming a halo ring. Bacillus thermoglucosidasius BtgMu1(KFCC-11085), which produces amylase, Bacillus thermoglucosidasius BtgcelMu1(KFCC-11084), which produces cellulase are characterized by repressing glucose consumption.
