PURPOSE: Provided is a preparation method of human Endostatin having angiogenesis inhibition activity. Thereby, human Endostatin of active form is mass-produced to inhibit proliferation of endothelial cell. CONSTITUTION: Provided are an expression vector, pRSETc-ENS containing a human endostatin gene and transformed E.coli, E.coli BL21(DE3)/pRSETc-ENS, therewith. A method of producing endostatin comprises the steps of: culturing transformants to prepare inclusion body of human endostatin; and refolding to prepare active human endostatin through by isolating the inclusion body, reducing using urea, beta-mercaptoethanol, and dithreitol(DTT) and dialyzing.
