New DNA encoding Arabidopsis thaliana 1-deoxy-D-xylulose-5-phosphate reductoisomerase for identifying modulators of the enzyme that can be used as herbicides, antibiotic or anti-malarial agents
A DNA (I) which encodes Arabidopsis 1-deoxy-D-xylulose-5-phosphate reductoisomerase (DXPR) or its functional fragment, is new. Independent claims are also included for the following: (1) a DNA having 80% homology to 2 specified 1732 base pair sequences, given in the specification; (2) a DNA or an RNA complementary to the DNA of (I) or (1); (3) an expression construct comprising the DNA of (I) or (1), and a sequence which is functionally linked and allows the DNA to be expressed; (4) a vector containing the DNA of (I), (1) or (3); (5) a host cell containing the expression construct of (3) or the vector of (4); (6) use of DXPR and/or 1-deoxy-D-xylulose-5-phosphate synthase (DXPS) modulators as herbicides, antibiotic agents or anti-malarial agents; (7) determining the activity of DXPR and/or DXPS synthase by: (a) converting pyruvate and glyceraldehyde-3-phosphate into or 1-deoxy-D-xylulose-5-phosphate with the aid of DXPS; and (b) converting the resulting 1-deoxy-D-xylulose-5-phosphate with the aid of DXPR; (8) identifying substances which modify the activity of DXPR or DXPS comprising (7) in the presence and in the absence of a test substance, and comparing the activities found in each case; (9) substances identified by the method of (8) except Fosmidomycin; and (10) a polypeptide comprising a sequence selected from two 477 amino acid sequences, given in the specification.
