A light beam (4) is deflected by a beam splitter (7) through an objective lens (5) on to a vessel containing the specimen (1). A focussing aid (9) focusses an image of itself on a boundary surface of the specimen which is recorded by the camera. The drive unit (14) moves the specimen in the z-direction so that successive boundary layers are traversed. Only at a boundary layer is the camera image sharp so that the distance between two sharp images gives the thickness of the intervening layer
