Extracting pure hepatoprotective agent phyllanthin in high yield from Phyllanthus amarus, by pulverizing and macerating dried leaves, percolating with organic solvent, defatting, chromatographing and crystallizing
Extraction and isolation of phyllanthin (I) from Phyllanthus amarus involves: (1) pulverizing dried leaves; (2) macerating with calcium carbonate; (3) percolating with a mixture of organic solvents ; (4) distilling off the solvents; (5) precipitating fats with a combination of organic solvents and filtering off the precipitate, and (6) separating and evaporating the n-hexane fractions. Extraction and isolation of phyllanthin (I) from Phyllanthus amarus comprises: (1) pulverizing the dried leaves; (2) macerating with calcium carbonate in a ratio of 19:1 to 7:3; (3) percolating with a mixture of organic solvents in a ratio of 9:1-1:1 for 10-30 hours at room temperature to 80 deg C; (4) distilling off the solvents; (5) precipitating fats with a combination of organic solvents and filtering off the precipitate; (6) separating and evaporating the n-hexane fractions; (7) chromatographing the obtained (I)-enriched residue on a silica gel column using solvents and solvent mixtures of increasing polarity; (8) combining the (I)-rich fractions; and (9) crystallizing pure (I) from n-hexane, cyclohexane or methanol.
