A method of purification of follicle stimulating hormone (FSH) from an FSH containing sample including the step of applying the sample in a first buffer to a dye affinity chromatography (DAC) matrix comprising a ligand dye, washing out contaminants from the chromatography matrix with a second buffer and subsequent elution of the FSH. The subsequent elution may be with a third buffer of 0.8 M NaCl or 1.0 M NaCl and include step wise increases in ionic strength. Alternatively the third buffer may have a pH greater than 8.0 or it may comprise a competitor of FSH binding to the dye ligand. Preferably the dye is Orange 1, Orange 2, Yellow 2 or Green 1. A further purification step by chromatography on a hydrophobic solid phase may also be used.
