The present invention discloses a unicellular regeneration process of plants of the specie (tequilana weber blue agave) by means of the in vitro culture technique of somatic indirect embryogenesis. It is about of plants generated from one single cell coming from the dedifferentiated tissues (callus); the process comprises the following steps: utilization of somatic cells which are taken to a culture mean that is prepared using the basic salts of the mean MS (Murashige and Soog, 1962), the vitamins L2 (Philips and Collins, 1979), gelled with agar and it is employed as a carbon source sucrose, and also supplemented with auxin 2, 4- dichlorophenoxyacetic acid and cytocinine benzyl-adenine, the exposure time to the induction mean is of about 15 and 80 days. Once the induction period is over, the embryogenic calluses are transferred to a solid culture mean for the expression and germination. The vegetal material is incubated under the following conditions: temperature of 27 +/- °C, a lumin ous intensity of 1500 lights and a photoperiod of 16 hours. This methodology is a tool which can be used to perform micro-propagation as well as the genetic enhancement in vitro of the specie.
