PURPOSE: A preparation method of a test strip for measuring glucose content in whole blood is provided, thereby effectively and cheaply measuring the glucose content in whole blood. CONSTITUTION: The test strip for measuring glucose content in whole blood comprises a matrix consisting of; enzyme solution which produces hydrogen peroxide from glucose component in whole blood sample passed through multiporous membrane, and oxygen; a pigment precursor forming pigments by the hydrogen peroxide and peroxidase; a polymeric material deposited thereon to support the enzyme solution and the pigment precursor.
