PURPOSE: Provided is a process for producing EPA ethylester and DHA ethylester economically in a high yield from a bio-oil component by using an affinity chromatography. CONSTITUTION: The process comprises the steps of: cool-precipitating a fatty acid ethylester mixture obtained by saponification or trans-esterification of the bio-oil component and performing a centrifugation or filtering to obtain a fraction containing an unsaturated fatty acid ethylester; dissolving the fraction containing the unsaturated fatty acid ethylester in an organic solvent, adding urea, heating, cool-precipitating, and filtering to refine firstly; reacting the firstly refined fraction and a monovalent metal ion solution to obtain a water-soluble fraction, adding sodium chloride and reacting, and extracting by the organic solvent to refine the unsaturated fatty acid ethylester secondly; applying the secondly refined unsaturated fatty acid ethylester to the affinity chromatography to obtain the EPA ethylester and the DHA ethylester in order. The organic solvent is n-hexane or diethylether and the monovalent metal ion is Ag+ or Cu+.
