PURPOSE: Provided are copper ion recovered superoxide(SOD) dismutase fusion protein which is efficiently transduced into the cells and a preparation method thereof, therefore harmful active oxygen species can be effectively removed from the human body by SOD. CONSTITUTION: The copper ion recovered superoxide(SOD) dismutase fusion protein having high cell penetration efficiency is produced by adding copper ion into an SOD fusion protein which is covalently bound with a protein transporting domain, wherein the protein transporting domain is any one selected from HIV-1 Tat(49-57), Tat(51-57), Tat(50-56), Tat(50-57), Tat(50-55), oligo-arginine consisting of 5 to 12 arginine residues, oligo-lysine consisting of 5 to 12 lysine residues, oligo(A+L) consisting of 5 to 12 arginine or lysine residues. The method for recovering copper ion into the SOD fusion protein comprises the steps of: purifying the SOD fusion protein which is covalently bound with the protein transporting domain; and adding copper ion into the purified SOD fusion protein.
