A polynucleotide encoding the nadA gene useful for the preparation of nicotinic acid or its derivatives, as probes for discovering RNA, cDNA and DNA to isolate polynucleotides or genes which code for quinolinate synthetase A
An isolated polynucleotide (I) from coryneform bacteria, comprising nucleotide sequence (seq) encoding for nadA gene, with a seq (S1) 70% identical to a seq. encoding polypeptide (pp) of 428 amino acid seq (S2) as given in the specification, a seq. (S3) encoding a pp with a seq 70% identical to (S2), a seq (S4) complementary to (S1) or (S3), or a seq with 15 bases of (S1), (S3), or (S4), is new. (I) from coryneform bacteria having a seq. encoding for nadA gene comprises: (a) a seq (S1) at least 70% identical to a polynucleotide which codes for a polypeptide which comprises (S2); (b) a seq (S3) which codes for a polypeptide which comprises a seq. which is at least 70% identical to (S2); (c) a seq (S4) which is complementary to (S1) or (S3); or (d) a seq comprising at least 15 successive bases of (S1), (S3) or (S4), the pp preferably has the activity of quinolinate synthetase A. Independent claims are also included for the following: (1) a coryneform bacterium (II) in which nadA gene is enhanced, in particular over-expressed; (2) a corynebacterium glutamicum DSM12455/pZ-nadAex (III) deposited on 25th October 2000 as DSM13794 at the Deutsche Sammlung fur Mikoorganismen und Zellkulturen (DSMZ = German Collection of Microorganisms and Cell cultures); and (3) a coryneform bacteria (IV) which contains a vector which contains (I).
