PURPOSE: To obtain a gene coing the subject enzyme obtained from a chromosomal DNA of Anabaena-variabilis belonging to Cyanophyta by cloning, useful for producing a transformed plant having improved fatty acid composition and resistant to low-temperature. ;CONSTITUTION: The gene coding a Δ9 unsaturation enzyme converting a saturated fatty acid bonded to a glycerolipid into an unsaturated fatty acid is produced by centrifuging a cultured liquid of Anabaena-variabilis belonging to Cyanophyta, adding an aqueous solution of sodium iodide to the collected cells, shaking the suspension, keeping at 37°C for 20min, collecting the cells by centrifuging, adding a buffer solution for DNA extraction and a lysozyme liquid, disintegrating the cell, recovering DNA by conventional method, subjecting the DNA to restriction enzyme treatment, linking to a phage vector, infecting to E.coli, screening by plaque hybridization with a probe, recovering DNA from positive plaque and subjecting the DNA to restriction enzyme treatment.;COPYRIGHT: (C)1995,JPO
