Method and reagent for detecting malaria-infected cells

PURPOSE:To simply inspect the presence/absence of infection with malaria by processing a liquid sample including a sample cell in a dyeing liquid composed of compounds of specific formulae, and optically detecting a cell dyed with fluorescence and infected with malaria. CONSTITUTION:A dyeing reagent is composed of a first dye of an auramine analogue represented by a formula I [R1-6 is a hydrogen atom, methyl radical or ethyl radical and X shows a halogen atom], a second dye of a condensed benzene derivative represented by a formula II, and a water-soluble organic solvent which is desirably ethylene glycol. The concentration of the first (second) dye of the dyeing reagent is preferably 0.005-0.4 (0.5) w/v%. In order to adjust the concentration of the dyes in a measuring solution, for example, a liquid sample is preliminarily diluted by a dilution liquid with a predetermined dilution ratio. The first dye is suitable to have the concentration of 1-80ppm. The measuring liquid prepared by mixing the dyeing reagent, liquid sample including a sample cell, and dilution liquid is measured by a flow cytometer. The presence of a cell infected with malaria is easily inspected in this manner.
