ISOLATION OF GROWTH HORMONE-RELEASING HORMOME RECEPTOR AND CHARACTERIZATION

PROBLEM TO BE SOLVED: To provide a method for isolation and purification of growth hormone- releasing hormone receptor. SOLUTION: The growth hormone-releasing hormone(GHRH) receptor bonding is characterized by using a specific bonding assay utilizing an iodized GHRM probe. A photoaffinity GHRH probe capable of photo-labeling and characterizing of the receptor is constructed. And a high affinity biotinylated GHRH analogue is constructed. A practically purified GHRH isolate is obtained by solubilization of GHRH-R/GHRH complex and extraction of the specifically bonded GHRH by using a mild surfactant and, next, affinity chromatography. The GHRH-R having sufficient purity for performing sequence determination of the receptor is produced by electrophoresis of the GHRH isolate.
