The present invention provides a monoclonal antibody that specifically reacts with a human growth hormone (20khGH) having a molecular weight of about 20,000 and does not substantially react with a human growth hormone (22khGH) with a molecular weight of about 22,000, and a method for producing the monoclonal antibody. The object of the present invention is to provide a cell line to be produced, a method for obtaining the cell line, and an immunoassay method of 20khGH using the monoclonal antibody. As a solution, 20khGH antibody-producing cells obtained from an animal immunized with 20khGH and Hybridoma cells are fused to produce anti-20khGH monoclonal antibodies. The anti-20khGH monoclonal antibody produced by this hybridomer is used to quantify 20khGH in blood by an enzyme immunoassay method. As a result, the monoclonal antibody of the present invention has extremely low cross reactivity to 22khGH and has a strong affinity for 20khGH. Therefore, the immunoassay method using the antibody enables the measurement of 20khGH at the baseline level of human blood, which is useful for clarifying the secretion dynamics of 20khGH.
