Allosterically regulatable hairpin ribozyme (I) is new. Independent claims are also included for: (1) polynucleotide (II) comprises a hairpin ribozyme having a substrate-binding domain A and a domain B, where catalytic activity is based on contact between A and B, and also, between A and B, a domain C which connects A and B and comprises a nucleic acid that binds a target molecule; (2) polynucleotide (IIa) similar to (II) but where B is a catalytic domain and C is a nucleic acid at least partly complementary to a sequence that binds a target molecule; (3) biosensor that comprises (I), (II) or (IIa); (4) method for identifying compounds that bind to a target molecule; and (5) kit for method (4) comprising (I), (II) or (IIa) and a signal-generating ribozyme substrate.
