Detecting nucleic acid-oligonucleotide hybridization events on a surface having built-in reference regions, is new. Detecting nucleic acid (NA) -oligonucleotide (ON) hybridization events comprises: (a) modifying a surface by binding at least two types of ligation oligonucleotides (ON1), with each type separated spatially; (b) adding to a sample that contains ligand oligonucleotides (ON2) another ligand oligonucleotide (ON3) that is a binding partner, with high association constant, for an ON1 present in a region (T100) on the surface and more of it is added than required for binding to all its specific ON1; (c) contacting sample and surface; (d) detecting hybridization; and (e) comparing values measured in (d) for the different types of ON1 with the result measured in T100. An Independent claim is also included for a kit for the new method comprising a modified surface and ON3.
