PPROBLEM TO BE SOLVED: To provide a preparation method and its system for testing BSE, which have such advantages that abnormal prions can be rapidly and accurately detected, and a lot of specimens can be treated. PSOLUTION: The preparation method is provided with a fist step in which cells of sampled parent specimens M are homogenized, a second step in which prescribed volumes of child specimens N are sampled from the homogenized parent specimens M, a third step in which a prescribed volume of enzyme material ( proteinase) K is injected and mixed as a reagent A, thereby decomposing proteins, a forth step in which heating is carried out at a temperature of a first level, a fifth step in which a prescribed volume of reagent B is added and mixed until solution blues, a sixth step in which a centrifugation process is carried out, and supernatant fluid is removed, a seventh step in which a prescribed volume of reagent CL is injected to carry out a condensing process, a eighth step in which heating is carried out at a temperature of a second level being relatively high, a ninth step in which a prescribed volume of diluent reagent D is injected and mixed, and a tenth step in which a sample for detecting the abnormal prions is made by dispensing a prescribed volume of the solution on a well of a microplate that adsorbs it.
