An isolated DNA encoding a CD40L mutein, selected from the group consisting of: (a) a DNA that encodes a polypeptide having an amino acid sequence as set forth in SEQ ID NO:2 wherein a cysteine at amino acid 194 is replaced with tryptophan and (b) a DNA that encodes a polypeptide fragment of SEQ ID NO:2, wherein the fragment comprises a polypeptide having the cysteine at amino acid 194 replaced with tryptophan and wherein the fragment retains the capacity to bind CD40.
