A plasmid useful as an internal control of a chain reaction of polymerase for the diagnosis of Agrobacterium tumefaciens, for which it is used for detection and quantification. The plasmid comprises a construction of DNA constituted by a fragment of a sequence of nucleotides corresponding to the tumor region of the pTi of Agrobacterium tumefaciens and in the interio of which there is inserted a fragment of a sequence of nucleotides corresponding to a second region of the pTi of Agrobacterium tumefaciens distinct from the first.
