A method for the production of biomass for the isolation of ccc plasmid DNA comprising (a) culturing a bacterial transformant in a bioreactor containing an antibiotic-free medium comprising (aa) a carbon source load; (Ab) a mixture of inorganic salts; (Ac) a nitrogen source; in batch culture conditions; (B) feeding able feedback to said culture of (a) at the end of the loading phase, after increasing the dissolved oxygen above a threshold-set point, a portion of a feed medium comprising (ba ) a carbon source; and (bb) a magnesium sa and (c) allowing the bacterial transformant to metabolize said feed means.
