A method of quantitating proteins in complex samples using capillaryelectrophoresis can be used both to determine the concentrationof a protein in a sample and to determine total protein concentration in thesample. In general, the method of determining the concentrationof a marker protein comprises: (1) adding a known quantity of an internalstandard compound to a sample containing at toast one protein,the internal standard compound selected from the group consisting of benzoicacid substituted with at least one halogen, producing adetector signal in relation to its concentration, and being capable ofelectrophoretic separation from the protein; (2) subjecting the sampleand the internal standard compound to capillary electrophoresis to separatethe protein and the internal standard compound from oath otherand from other components in the sample; (3) measuring the detector signalproduced by the internal standard compound and a detectorsignal produced by the protein to determine a ratio of protein signal tointernal standard signal; and (4) determining the concentration ofthe protein in the sample from a standard curve of protein concentrationversus the ratio of protein signal to internal standard signal. Atypical protein measurable by the method is albumin. Typically, measurementsare made at 214 em and the internal standard compound is2,4-dichlorobenzoic acid
