IMMOBILIZED G-PROTEIN COUPLED RECEPTORS ON LIQUID CHROMATOGRAPHY STATIONARY PHASES AND USE THEREOF FOR ON-LINE SCREENING

This invention provides an immobilized G-protein coupled receptor(GPCR) on a support in a liquid chromatography system. The method of theinvention provides means of evaluating the attachment of an agent to a GPCRcomprising the steps of (a) immobilizing a GPCR on an artificial membranesupport in a column, (b) exposing the support with the GPCR to test the agentatvarying concentrations in a liquid chromatography system, (c) eluting the testagentfrom the column, and (d) evaluating the elution profile of the test materialfrom thecolumn. Using this method, it is possible to evaluate the interaction of thetestagent with the GPCR. Following elution, it is possible to directly determinemolecular structure by passing the elute through other testing devices such asamass spectrometer.
