It is intended to provide nerve degenerative disease neurons within a short time by differentiating disease mutation knockin ES cells by a highly efficient differentiation method. A knockin ES cell having a gene mutation, which is causative of Alzheimer’s disease (i.e., a nerve degenerative disease), is differentiated by a novel highly efficient differentiation method to thereby establish Alzheimer’s disease model neurons. Thus, a knockin ES cell having a gene mutation causative of Alzheimer’s disease is established. Further, the ES cell is differentiated by the novel highly efficient differentiation method and thus Alzheimer’s disease model neurons expressing a neuron marker at a ratio of about 90% or higher can be established. Furthermore, a disease gene mutation knockin animal is constructed and thus a disease model animal, in particular, an Alzheimer’s disease model is provided.
