PURPOSE: A method for regulating tapasin-dependency of MHC class I molecule is provided, thereby regulating the cell surface expression of a specific histocompatibility antigen, and treating several symptoms associated with the tapasin-dependency of MHC class I molecules. CONSTITUTION: A mutant MHC class I molecule is characterized in that an amino acid residue at position 114 in wild MHC class I molecule is substituted from the basic residue to the acidic residue or from the acidic residue to the basic residue, wherein the basic residue is histidine, arginine or lysine and the acidic residue is aspartic acid or glutamic acid. A recombinant expression vector contains the gene encoding the mutant MHC class I molecule. A method for regulating tapasin-dependency of MHC class I molecules comprises substituting the amino acid residue at position 114 from the basic residue to the acidic residue or from the acidic residue to the basic residue.
