PURPOSE: Provided is a method for increasing the transformation efficiency of soybeans by pretreating damaged beans with AAM medium containing hormones BA and TDZ(thidiazuron), followed by inoculation with Agrobacterium LBA4404. The method is useful for molecular breeding industry. CONSTITUTION: The method for increasing transformation efficiency of soybeans comprises the steps of: germinating soybeans; culturing Agrobacterium LBA4404 containing a plasmid pTOK233; preparing AAM medium containing hormones BA and TDZ(thidiazuron); diluting Agrobacterium LBA4404(pTOK233) with the AAM medium and infecting the damaged cotyledon of the soybeans with the diluted medium; transferring the cotyledon into a shoot inducing medium and culturing it at 24 deg. C for 3 days in a dark room to form shoots or calluses of the soybean; and analyzing the transformation efficiency of the soybeans by GUS chromogenic reaction, wherein the amount of BA is 5 to 10 ppm; and the amount of TDZ is 2 to 5 ppm.
