Detecting effect of substances on beta-cell function and mass, for identifying agents for treating late stages of diabetes type II, comprises immunoblotting and mitogen-activated protein kinase and pull-down assays
Method for detecting the effect of substances on the function and mass of insulin-producing beta cells (A). Method for detecting the effect of substances on the function and mass of insulin-producing beta cells (A) comprises: (1) preparing and culturing human islet cells; (2) transforming the cells with Elk1 or CREB (cyclic adenosine monophosphate response element binder) transactivator domain and a reporter gene (RG); (3) stimulating the cells with glucose and a test compound (I); (4) measuring activity of the reporter; (5) performing immunoblotting and mitogen-activated protein kinase (MAPK) and pull-down assays; and (6) evaluating the effect of (I) by comparing results with those from positive and negative controls.
