The milk (1) sample being tested and its progesterone (4) are incubated with the anti-progesterone (2) antibody. It is placed in direct competition with a progesterone conjugated with an enzyme (5). Development produces a color by addition of a substrate (7) for the enzyme. A colorimetric reading is taken, using a fixed threshold in the colorimeter.
