This invention provides a method of isolating sexspecific DNA sequences in fish that includes the steps ofhybridizing an excess of denatured, sheared DNA of afirst sex with a lesser amount of denatured DNA of asecond sex that has been digested with a restrictionendonuclease that produces sticky ends, and ligatingresulting double-stranded fragments having two stickyends derived from the DNA of the second sex into acloning vector. This invention also provides DNA probesisolated by the preceding method. This invention alsoprovides methods of identifying sex specific DNAsequences using the aforementioned probes or, by usingPCR primers derived from the aforementioned probes.
