Compositions and methods for achieving therapeutic treatment of diseases andfor regulating gene expression in biological experimentalsystems via interference with 5' capping of targeted nascent mRNAs aredisclosed. Interference with capping is achieved in accordance withpreferred embodiments utilizing antisense oligonucleotide compounds which arecomplementary to the targeted nascent RNA and preventbinding of capping enzyme either via steric interference or by chemicalmodification of the nascent mRNA.
