PURPOSE: DNA-binding proteins and gene expression systems using the same are provided, therefore the DNA-binding proteins and the gene expression systems can be useful for regulation of gene expression or screening of an antagonist for a specific ligand. CONSTITUTION: The DNA-binding proteins comprises a DNA-binding domain of purine repressor(DBD), a hinge region of purine repressor and a dimerization domain(DD) of Gal4, wherein the DNA-binding domain and the hinge region are composed of from amino acids 1 to 54 in the amino acid sequence of E. coli purine repressor, and the dimerization domain is composed of from amino acids 42 to 148 in the amino acid sequence of Gal4 protein. The DNA-binding protein has the amino acid sequence of SEQ ID NO: 4. A transcription factor comprises the DNA-binding domain of purine repressor(DBD), the hinge region of purine repressor, the dimerization domain(DD) of Gal4, a first linker and a transcription activating domain, wherein the transcription activating domain is derived from VP16, p65 of NF-kappaB or SREBP1a.
