A raster microscopy procedure takes a pre image of a sample with areas of different fluorescent gain and automatically determines the different areas and appropriate illumination intensities and detection sensitivity for the final image. Independent claims are included for a raster microscope using the procedure. Manual marking of areas on the display is also possible.
