In vitro detection of a systemic inflammatory response syndrome (SIRS) comprises detecting abnormal expression of disease-related genes. Method for in vitro detection of systemic inflammatory response syndrome (SIRS) comprises: (1) isolating sample RNA from a mammal; (2) labeling the RNA and/or at least one DNA that is a SIRS-specific gene (or its fragment) with a detectable marker (M); (3) contacting sample RNA and the DNA under hybridization conditions; (4) contacting control RNA (representing a non-pathological state) with the DNA under similar conditions; (5) quantitative detection of the signals from (M); and (6) comparing the signals to determine if SIRS-specific genes are expressed more strongly or more weakly in the sample, relative to control. Independent claims are also included for the following: (1) similar method for in vitro detection of (severe) sepsis or sepsis-like conditions, where the DNA used is a gene (or fragment) specific for these diseases; (2) similar method for in vitro detection of SIRS, (severe) sepsis or sepsis-like conditions based on isolation and labeling of peptides, reaction with antibodies (Ab) specific for these diseases and comparing levels of peptides with those found in healthy controls; and (3) use of recombinant or synthetic nucleic acid (partial) sequences, or derived proteins or peptides, alone or in mixtures, as calibrants in assays for the specified diseases, for evaluating activity or toxicity in screening for active agents, and/or for preparation of agents for treatment or prevention of the specified diseases.
