Bisulfite conversion (M1) of DNA, involves reacting genomic DNA with a bisulfite reagent, where the reaction is carried out in the presence of a compound out of the group of dioxane, one of its derivatives and a similar aliphatic cyclic ether. Bisulfite conversion (M1) of DNA, involves reacting genomic DNA with a bisulfite reagent, where the reaction is carried out in the presence of (a) a compound out of the group of dioxane, one of its derivatives and a similar aliphatic cyclic ether, (b) a compound of formula called (1), or (c) the reaction is conducted at a temperature in the range of 0-80 [deg]C and that the reaction temperature is increased to a range of 85-100 [deg]C briefly during the course of the conversion (thermospike). An independent claim is also included for a kit, comprising a reagent containing bisulfite, denaturing reagents or solvents, and scavengers and primers for the production of amplificates, and optionally an ultrafiltration tube or instructions for performing (M1).
