Stable anionic liposome depot system for prolonged release of protein or peptide drugs, e.g. insulin, comprising phosphatidyl choline, cholesterol, anionic lipid(s) and cationic polymer
The use of a depot system (I) containing anionic liposomes is claimed for retarded drug release, where (I) comprises protein and/or peptide active agent(s), saturated synthetic phosphatidyl choline(s) (PC's), cholesterol, anionic lipid(s), a cationic polymer. The use of a depot system (I) containing anionic liposomes is claimed for retarded drug release, where (I) comprises: (a) protein and/or peptide active agent(s); (b) saturated synthetic phosphatidyl choline(s) (PC's) selected from dimyristoyl, dipalmitoyl and distearoyl PC's; (c) cholesterol at 35-50 mol. %; (d) anionic lipid(s) selected from dimyristoyl, dipalmitoyl and distearoyl phosphatidyl glycerols (PG's), dimyristoyl and dipalmitoyl phosphatidyl serines (PS's) and cholesterol hemisuccinate, at 5-20 mol. % of the liposome membrane; and (e) a cationic polymer. Independent claims are included for the use of depot systems as above for administration of certain active agents other than proteins and/or peptides.
