By manipulating an interrupt codon in HA1, the expression of HA1 is ensured. Liposome DNA vaccine encapsulation and intranasal inoculation is sufficient to elicit protective immunity at a significantly lower dosage compared to a DNA vaccine expressing the full length HA gene. Lower dosage reduces the risk of anti-DNA antibody induction. Intranasal inoculation directly into respiratory epithelial cells reduces the risk of DNA integration. The vaccine according to the invention is advantageous over current inactivated or live attenuated vaccines, because updating the vaccine only requires replacing the coding sequence with the new virus.
