A grafted antibody, or a functional fragment, comprises one or more complementarity determining regions (CDRS) that has a least one amino acid substitution in one or more CDRs of a heavy or light chain CDR selected from a specific group. The grafted antibody has specific binding activity for a cryptic collagen epitope. The antibody can be used to inhibit angiogenesis, tumour growth and metastasis.
