A novel method of constructing a self-assembly of probes which comprises preliminarily preparing a plural number of dimer probes and then increasing the types, thereby enabling to shorten the reaction time of the construction of the self-assembly compared with the conventional cases and increase probe regions usable in detecting a target gene. A plural number of systems each containing a pair of probes for forming a dimer, in which each of a pair of oligonucleotides is divided into three regions (i.e., a 3"-terminal region, a central region and a 5"-terminal region) and the base sequences in the central regions of the oligonucleotides are made complementary to each other while the base sequences of the 3"-terminal and 5"-terminal regions are made non-complementary to each other, are formed. Then a plural number of pairs of probes for forming a dimer in these systems are hybridized to thereby allow the oligonucleotides to form a double-stranded self-assembly.
