Simultaneous generation of time-resolved fluorescence images and emission spectra, for determining the parameters of samples in multi-well in vitro fluorescence assays, and in DNA chips, comprises: (a) directing a laser beam to a fluorescent sample; (b) splitting the emitted light into two; and (c) combining the two beams in a multiparameter acquisition system of two ZOKEPZ-based detectors. Simultaneous generation of time-resolved fluorescence images (fluorescent lifetime imaging) and time-resolved emission spectra, also emission anisotropy patterns, based on time- and space-correlated single-photon counting (ZOKEPZ), for determining the parameters of samples such as live cells in multi-well in vitro fluorescence assays and in DNA chips, comprising: (a) directing a pulsed, high frequency, polarized laser beam to a fluorescent sample; (b) splitting the emitted light into two beams (T1 and T2) using a beam splitter; and (c) combining the two beams in a multiparameter acquisition system of at least two ZOKEPZ-based detectors. For maximum recovery of information, all physical parameters of each individual photon are detected, then stored and evaluated in control electronics.
