VECTOR AND METHOD FOR IMPROVING STABILITY OF GENE TRANSFECTED INTO MAMMALIAN CELL

PPROBLEM TO BE SOLVED: To provide a method available for a variety of mammalian cells as a host and for stabilizing a transfected target gene. PSOLUTION: Disclosed is a vector for stabilizing a gene transfected into mammalian cells, wherein a replication fork from the replication initiation region of the vector and transcription from an arbitrary gene incorporated into the vector have the same orientational structure. The vector has a structure comprising a sequence inhibiting the replication fork from the replication initiation region or a poly(A) addition sequence in a region in which the replication fork from the replication initiation region of the vector collides head-on with the transcription from the arbitrary gene incorporated into the vector. The vector has a structure free from a nuclear matrix binding region in the region in which the replication fork from the replication initiation region of the vector collides head-on with the transcription from the arbitrary gene incorporated into the vector.
