PPROBLEM TO BE SOLVED: To provide a nerve cell death-promoting factor or its derivative, to provide a method for measuring the same, and to provide a technique related to the nerve cell death-promoting factor, including use of the same. PSOLUTION: A noncleavable proBDNF derivative is provided, wherein the derivative comprises (a) a protein comprising a specific amino acid sequence or (b) a protein comprising an amino acid sequence in which one, two, or more amino acids in the specified amino acid sequence are subjected to substitution, addition, deletion, or insertion and having promoting activity of nerve cell death and/or fiber dropout of a cholinergic neuron.
